Abstract

  Stable TLR-transfected cell lines for synthetic TLR agonists detection was generated. Unfortunately, the sensitivity was not sufficient to detect endogenous TLR agonists from pathogens making these cell lines not suitable for IVD. A TLR3 agonist has been developped up to the lead. Mechanisms of TLR pathways downregulation by HPV have been deciphered. TLR3 expression level correlates with the stage of HPV infection in precancerous cervical lesions. TLR3 but also TLR7 agonists efficacy alone or in combination with lapatinib has been studied in tumor models.  


Objectives

  Set up of new cellular assays based on TLR mediated recognition for the detection and partial diagnostic of inapparent microorganisms. Rational development of synthetic TLR agonists, especially TLR3 agonists, based on the use of these cellular assays Identification of therapeutic indications in the field of viral infections or viral induced cancer for the treatment by TLR synthetic agonists, especially TLR3 agonists.
